宫瘤消胶囊对子宫内膜异位症大鼠异位内膜中Bcl-2、Bax蛋白表达的影响

摘    要:目的:观察宫瘤消胶囊对子宫内膜异位症(内异症)模型大鼠异位内膜中凋亡基因Bcl-2、Bax蛋白的表达的影响,为该药治疗内异症提供理论依据。方法:于大鼠动情期行自体移植法建立子宫内膜异位症大鼠模型,造模成功后随机分为模型组、桂枝茯苓胶囊组及宫瘤消胶囊小、中、大剂量组,各组分别给予指定药物进行灌胃干预,另取一组正常大鼠为正常内膜组,用生理盐水灌胃。用免疫组织化学法检测各组大鼠异位内膜中凋亡基因Bcl-2和Bax的蛋白表达情况。结果:与正常内膜组比较,模型组Bax蛋白表达明显降低,Bcl-2蛋白的表达明显升高,Bax/Bcl-2比值降低(P0.01);与模型组比较,宫瘤消胶囊各剂量组Bax蛋白表达明显升高,Bcl-2蛋白表达明显降低,Bax/Bcl-2比值增加(P0.05,P0.01),且作用随药物剂量增加而增加(P0.05,P0.01)。结论:宫瘤消胶囊可能通过调节凋亡基因Bcl-2和Bax的表达来实现对异位内膜的凋亡的控制。

Abstract:  Objective:To investigate the effect of Gongliuxiao capsule on the expression of Bcl-2 and Bax in ectopic endometrium of endometriosis rats, and provide theoretical basis for the treatment of endometriosis with Gongliuxiao capsule.   Methods: Rat model of endometriosis was established by autologous grafting operation method at estrous time. Successful modeling rats were randomly divided into the model group,Guizhi Fuling capsule group,Gongliuxiao capsule groups with low-,middle-and high-dose. Each group was orally administrated with the designated drugs. Another group of normal rats was set as normal endometrium group which treated with normal saline. The expression of apoptosis gene Bcl-2 and Bax in ectopic endometrium of rats were detected by immunohistochemistry.   Results: Compared with the normal endometrium group, the expression of Bax in the model group was significantly decreased ,and the expression of Bcl-2 was significantly increased, the ratio of Bax and Bcl-2 was decreased (P<0.01); compared with the model group , the expression of Bax was significantly increased in the Gongliuxiao capsule groups with different doses, the expression of Bcl-2 was significantly decreased, the ratio of Bax and Bcl-2 was increased (P<0.05, P<0.01), and these effects showed in a dose-dependent manner(P<0.05,P<0.01).   Conclusion: Gongliuxiao capsule could control the apoptosis of ectopic endometrium by regulating the expression of Bcl-2 and Bax.
