松果菊苷对过氧化氢损伤的PC12细胞的保护作用及机制研究

摘    要:目的研究松果菊苷(echinacoside,ECH)对过氧化氢(H2O2)诱导的PC12细胞损伤的保护作用及机制。方法用终浓度为0.4 mmol.L-1的H2O2损伤PC12细胞,测定细胞存活率和Na+,K+-ATP酶的活性;用激光共聚焦法(LSCM)检测线粒体膜电位(MMP);用RT-PCR法检测Bcl-2和p53 mRNA表达量的变化。结果10 mg.L-1松果菊苷可以提高H2O2损伤的PC12细胞存活率和Na+,K+-ATP酶的活力、升高线粒体膜电位、降低p53 mRNA水平但增高Bcl-2 mRNA水平(P<0.05或P<0.01)。结论松果菊苷可保护H2O2诱导的PC12细胞损伤,升高MMP、下调p53mRNA和上调Bcl-2 mRNA可能是其作用机制。

Abstract:Aim To investigate the protective effect and mechanisms of echinacoside(ECH) on H2O2-injured PC12 cells.Methods After PC12 cells were treated with 0.4 mmol·L-1 H2O2 and different concentrations of ECH,the cell viability was measured by MTT method,the mitochondrial membrance potential(MMP) was evaluated under LSCM and the expressions of Bcl-2,p53 mRNA were detected by RT-PCR.Results Cell viability and Na+,K+-ATP ase were significantly elevated by ECH from 27.9% to 51.0%,from 5.39 mmolPi·g-1Pro·h-1 to 6.84 mmolPi·g-1Pro·h-1,respectively.MMP was significantly heightened by ECH through judging the change of JC-1 R0/Rt from 1.57 to 1.22.Meanwhile,the level of p53 mRNA was decraesed and that of Bcl-2 mRNA increased(P<0.05 or P<0.01).Conclusions ECH can protect PC12 cells from H2O2-induced damage through heightening MMP,downregulating the expression of p53 mRNA but upregulating the expression of Bcl-2 mRNA.
