摘    要:目的探讨表皮生长因子受体阻断剂AG1478对肾癌粒细胞生成素反应细胞1(granulopoietinresponsivecell1,GRC1)的诱导凋亡作用及其作用机制,为肾癌的生物学特征研究和药物治疗提供理论依据。方法以不同浓度的AG1478作用于体外培养的肾癌GRC1细胞,噻唑蓝(MTT)法检测细胞生长抑制率,应用流式细胞仪观察细胞周期的改变,应用DNA电泳及电子显微镜观察细胞凋亡。结果AG1478抑制肾癌GRC1细胞的增殖,诱导细胞周期停留在G1期,诱导肾癌GRC1细胞凋亡。结论表皮生长因子受体特异性阻断剂可抑制肾肿瘤细胞的增殖,有望成为抗肾脏恶性肿瘤药物。

Tyrophostin AG1478 to induce granulopoietin responsive cell-1 apoptosis and its mechanism
Abstract:Objective AG1478,an epidermal growth factor receptor blocker,shows an apoptosis induction to granulopoietin responsive cell-1 (GRC-1) of renal cancer,the paper is to survey its mechanism and afford a theoretical basis for studying the biological feature and drug therapy of renal cancer.Methods Different concentrations of AG1478 were used to observe its inhibitory effect on in vitro cultivated GRC-1;the cell growth inhibition was checked by MMT method,the cell cycle change was examined by flow cytometry,and the cell apoptosis was observed by DNA electrophoresis and electron microscopy.Results AG1478 inhibited GRC-1′s multiplication,made the cell cycle stay at its presynthetic gap (G_1),and induced GRC-1 of renal cancer to apoptose.Conclusion AG1478,a specific blocker of epidermal growth factor receptor,is able to inhibit the multiplication of renal tumor cells,it is a promising drug for treating renal malignancies.
