水蛭提取液对视网膜色素上皮细胞内游离钙离子的影响

摘    要:目的观察水蛭提取液对视网膜色素上皮细胞(retinal pigment epithelium,RPE)内游离钙离子(Intracellular liber calcium ion,Ca2 ]i)浓度的影响。方法利用体外培养的视网膜色素上皮细胞,采用双波长荧光分光光度计测量法测定视网膜色素上皮细胞内Ca2 ]i浓度。结果空白组(A)133.4517±13.17789、凝血酶组(A1)192.2037±4.54292、水蛭组(A2)96.9935±8.80148、水蛭和凝血酶同时加组(A3)142.0867±26.47195、先加水蛭后加凝血酶组(A4)162.7284±29.13950、先加凝血酶后加水蛭组(A5)130.4810±20.48673。结论水蛭提取液能使RPE细胞内游离Ca2 浓度下降,为进一步深入研究水蛭提取液抑制RPE细胞增殖的作用机制,防治增生性玻璃体视网膜病变(proliferative vitreo-retinopathy,PVR)提供了实验室依据。

Abstract:OBJECTIVE To observe effect of percolation leech extract on intracellular liber calcium ionCa~(2 )]i of retinal pigment epithelium(RPE)cultured in vitro.METHODS Use retinal pigment epithelium(RPE)culture in vitro,to observe effect of leech extract on retinal pigment epithelium(RPE)Ca~(2 )]i.RESULTS One hundred and twenty-eight mg/ml leech extract can reduceCa~(2 )]i of retinal pigment epithelium(RPE)cultured in vitro.CONCLUSIONS Leech extract can reduceCa~(2 )]i of retinal pigment epithelium(RPE)cultured in vitro to provide experimental evidence for drug treatment on proliferative vitreoretinopathy(PVR).
