法舒地尔对去势大鼠阴道平滑肌RhoA/Rho激酶的影响

目的:研究法舒地尔对去势大鼠阴道平滑肌RhoA/Rho激酶的影响。方法:30只10周龄雌性SD大鼠随机均分成:对照4周组(A组)、对照6周组(B组)、去势4周组(C组)、去势6周组(D组)、去势+法舒地尔治疗4周组(E组)和去势+法舒地尔治疗6周组(F组)。法舒地尔治疗4、6周测定各组大鼠血清雌二醇(E2)水平,并取阴道平滑肌组织测定ROCKⅠ、ROCKⅡ的表达。结果:血清E2水平C组、E组较A组显著降低(P<0.01),D组、F组较B组显著降低(P<0.01),C、D、E、F组之间无统计学意义(P>0.05)。阴道平滑肌细胞ROCKⅠ、ROCKⅡ的表达C组较A组、E组显著升高(P<0.05),D组较B组、F组显著升高(P<0.05),A、B、E、F组之间无统计学意义(P>0.05)。结论:法舒地尔可抑制去势大鼠阴道平滑肌RhoA/Rho激酶高表达。

Effect of fasudil on expression of RhoA/Rho-kinase in vaginal smooth muscle cells of castrated rats

Objective: To determine the effect of fasudil on the expression of RhoA/Rho-kinase(ROCK)in vaginal smooth muscle cells of castrated rats.Methods: 30 female Sprage-Dawley rats of 10 weeks were randomly divided into group A(4-week control group),B(6-week control group),C(4-week bilateral ovariectomy),D(6-week bilateral ovariectomy),E(4-week bilateral ovariectomy plus treatment with fasudil) and F(6-week bilateral ovariectomy plus treatment with fasudil).4 and 6 weeks after fasudil treatment,the levels of serum estradiol were determined and the expressions of RockⅠ and RockⅡ in the vaginal smooth muscle were detected by Western blot.Results: The levels of serum estradiol were significantly lower in group C and group E than in group A(P< 0.01) and in group D and group F than in group B(P<0.01).There were no significant differences among group C,group D,group E and group F(P>0.05).The expressions of RockⅠ and RockⅡ were significantly higher in group C than in group A and group E(P<0.05),and in group D than in group B and group F(P<0.05).There were no significant differences among group A,group B,group E and group F(P >0.05).Conclusion: Fasudil may suppresses the high-expression of RhoA/Rho-kinase in vaginal smooth muscle cells of castrated rats.
