丝素纤维与海马神经元的生物相容性细胞培养研究

摘    要:目的:研究丝素与体外培养的大鼠海马神经元的生物相容性。方法:采用制备丝素与SD大鼠胎鼠海马神经元共培养7天,每天倒置显微镜观察,免疫细胞化学染色激光共聚焦显微镜观察、分析。结果:通过光镜观察,发现大鼠海马神经元与丝素纤维共培养3天后,海马神经元附着于丝素纤维,神经突起沿丝素纤维向远处生长、延伸;培养7天后海马神经元形成神经元网络,包绕丝素纤维。免疫细胞化学染色显示附着于丝素上的细胞为神经元。结论:海马神经元沿着丝素纤维包裹生长,提示丝素纤维与海马神经元具有良好的生物相容性。

Abstract:Objective: To explore the biocompatibility of silk fibroin with co-culture hippocampal neurons.Methods: Silk fibroin were prepared before co-culture with hippocampal neurons,then light and laser confocal microscopy were conducted with the method of immunocytochemistry.Results: The cells clasped around silk fibroin after 3d,co-culture and the cells wrapped up the silk fibroin filaments after 7d co-culture.The stretched cells on the silk fibroin are neurons that were identified by immunocytochemistry.Conclusions: Neuritis of hippocampal neurons can extend along silk fibroin filaments,the results suggested that silk fibroin has good biocompatibility with hippocampal neurons.
