TRIM28在乳腺癌中的表达及临床价值

摘    要:
目的:分析TRIM28在乳腺癌中的表达及其预后价值。方法:乳腺癌患者114例运用荧光实时定量PCR及免疫组织化学法检测癌组织中TRIM28的表达,分析TRIM28表达与患者临床病理特征的关系,使用Kaplan-Meier法和Log-Rank检验分析总生存期和无进展生存期,使用Cox回归模型分析影响预后的危险因素。结果:乳腺癌组织中TRIM28表达水平显著高于癌旁组织,差异有统计学意义(P<0.001)。TRIM28表达与乳腺癌淋巴结转移(P=0.013)、TNM分期(P=0.013)、分子分型(P=0.014)密切相关。TNM分期等级越高,癌组织TRIM28表达水平越高,差异有统计学意义(P<0.001)。乳腺癌组织免疫组化评分高于癌旁组织,差异有统计学意义(P<0.001)。三阴性乳腺癌免疫组化评分高于非三阴性乳腺癌,差异有统计学意义(P=0.01)。与阴性者相比,TRIM28阳性表达患者总生存期(P=0.001)和无进展生存期(P<0.001)更短,差异均有统计学意义。多因素分析显示,分子分型、TRIM28表达是影响总生存期的独立危险因子,TRIM28、HER-2表达、分子分型是影响无病生存期的独立危险因子。结论:TRIM28在乳腺癌中高表达,是预测乳腺癌不良预后的潜在生物标志物,有望作为乳腺癌治疗的新靶点。

Abstract:
Objective:To analyze the expression of tripartite motif containing 28(TRIM28) in breast cancer, and to investigate its role in promoting the progression and predicting the prognosis in breast cancer. Methods:TRIM28 expression in breast tissues was detected by quantitative real-time PCR(qRT-PCR) assay and immunohistochemistry. The association between TRIM28 expression and clinicopathological characteristics was analyzed by chi-square test. The overall survival and recurrence rates were evaluated by the Kaplan-Meier method and compared with the log-rank test. Univariate and multivariate analyses were performed by using the Cox regression method. Results:TRIM28 expression was significantly higher in breast cancer tissues. Observably, TRIM28-positive staining was associated with nodal status(P=0.013), TNM classification (P=0.013), molecular subtype (P=0.014). Additionally, high TRIM28 expression is an independent prognostic factor for poor overall survival(OS) and progression-free survival(PFS). Conclusions:TRIM28 is overexpressed in breast cancer, and is relevant to clinicopathological characteristics. It also promotes progression of breast cancer and predicts poor prognosis. Therefore, TRIM28 could be a prognostic biomarker and novel therapeutic target in breast cancer.
