五味子醇甲抑制6-羟基多巴胺诱导PC12细胞凋亡的研究

摘    要:目的 研究五味子醇甲(Schizandrin,SCH)的神经保护作用并探索其作用机制。方法 以PC12细胞为模型细胞,采用四氮唑(MTT)比色法检测6-羟基多巴胺(6—0HDA)对PC12细胞活性的影响;采用放射免疫法测定PC12细胞对培养液中^3H—D,L-谷氨酸的摄取;采用高效液相色谱法(HPLC)测定细胞外液中谷氨酸的浓度。结果 6—OHDA能抑制PC12细胞摄取谷氨酸,提高胞外谷氨酸的浓度,降低细胞的存活率;SCH能增强PC12细胞对谷氨酸的摄取,降低胞外谷氨酸的浓度,并拮抗6-OHDA对PC12细胞摄取谷氨酸的抑制作用和对细胞存活率的影响。结论 SCH对PC12细胞有保护作用,其保护作用机制可能与增强谷氨酸转运体(Glutamate transporters,GluTs)的功能有关

Study on the Protective Effects of Schizandrin on 6-OHDA-induced Apoptosis of Cultured PC12 Cells

Abstract:OBJECTIVE To study the protective effects and mechanisms of Schizandrin(SCH) on 6-OHDA-induced PC12 cells apoptosis. METHODS The viability of PC12 cells was investigated by MTT method; The glutamate uptake into PC12 cells was measured by using radioimmunoassay method; The extracellular glutamate levels of PC12 cells was detected with HPLC. RESULTS Treatment with 6-OHDA produced inhibitory effect on viability and glutamate uptake into cells. Incubation with SCH caused increase in the viability and glutamate uptake into PC12 cells, which completely reversed 6-OHDA-induced uptake inhibition. CONCLUSION SCH may modulate glutamate transporters and reduce extracellular glutamate levels, through which it protects PC12 cells against 6-OHDA -induced injury and may be useful in the treatment of PD.
