摘    要:目的:用二甲基亚硝胺(Dimethylnitrosamine,DMN)造模对肝纤维化发生机制进行探讨,为临床诊断和治疗提供理论依据。方法:采用电镜技术、免疫组织化学技术对肝纤维化形成过程中肝星状细胞的变化进行动态观察,并观察DMN造模后7、14、21、28天的肝重、体重及肝体比。结果:肝纤维化模型大鼠肝星状细胞可被激活、转化为成纤维细胞。随造模时间延长,大鼠肝重在注射DMN后第7天有明显的增加,随后逐渐减轻,到第28天达到最小。结论:肝星状细胞激活、转化在肝纤维化发生机制中起重要作用。

Abstract:Objective:To study the mechanisms of hepatic fibrosis induced by Dimethylnitrosamine(DMN) in rats and provide theoretical bases for clinical diagnosis.Methods: Electron microscope and immunohistochemical technique were used to dynamically observe the changes of hepatic satellite cells.And the hepatic weight,body weight were also observed on 7th,14th,21st and 28th day respectively after DMN was used.Results: In hepatic fibrosis model rats,the satellite cells could be activated and transformed into fibroblast.The body weight of rats obviously increased at 7th after DMN treatment,decreased after that,and got to minimum at 28th day.Conclusions: Activation and transforming of satellite cells has important effects in the development of hepatic fibrosis.
