替尼泊甙联合其他抗肿瘤药对BGC-823细胞体外增殖的影响

目的:比较替尼泊甙(VM-26)与紫杉醇(TAX)、顺铂(DDP)、5-氟尿嘧啶(5-FU)联合用药对胃癌细胞BGC-823体外生长的抑制作用。方法:MTT法测定VM-26单药及分别与TAX、DDP、5-FU联合用药对胃癌细胞BGC-823的细胞生长抑制率,计算联合用药指数。结果:单药作用下BGC-823细胞生长抑制率随药物剂量的增加而增大,TAX、VM-26、DDP、5-FU的IC50依次增大。VM-26分别和TAX、DDP、5-FU联合应用时,BGC-823细胞生长抑制率较相同剂量单药应用时增大,IC50降低;联合用药指数分别为1.22,0.43,1.04。结论:VM-26可抑制胃癌细胞的生长,VM-26与DDP联合有协同作用,与5-FU联合有相加作用,与TAX联合有拮抗作用。

Effects of teniposide combined with other anticancer drugs on proliferation of BGC-823 cell lines in vitro

Aim:To investigate the effects of teniposide(VM-26)combined with TAX,DDP,and 5-FU,respectively,on proliferation of gastric cancer cell line BGC-823.Methods:The growth inhibition rate of BGC-823 cells treated with VM-26 combined with TAX,DDP,5-FU,respectively,were examined by MTT assay,and combined medication index(CI)was calculated.Results:The growth inhibition rates of BGC-823 cells treated with single medicine increased in a dose-dependent manner,and IC50 of TAX,VM-26,DDP,5-FU increased in turn.Compared with those of single medicine treatment,the inhibition rates of BGC-823 cells treated with VM-26 combined with TAX,DDP,and 5-FU,respectively,were higher,and IC50were lower,CI was 1.22,0.43,1.04,respectively.Conclusion:VM-26 could significantly inhibit the proliferation of gastric carcinoma cell in vitro.VM-26 combined with DDP,TAX,and 5-FU show synergic,antagonistic,and additivity action,respectively.
