组织芯片技术在研究肿瘤组织中抗原表达的应用

摘    要:目的:研究多种肿瘤组织芯片、肿瘤进展组织芯片以及肿瘤预后相关组织芯片中多种抗原的表达,明确组织芯片技术在肿瘤研究中的应用。方法:运用组织芯片技术和免疫组织化学法检测多种肿瘤组织中雌激素受体(ER)、孕激素受体(PR)的表达、肺癌进展组织中P53的表达及肺鳞癌组织中P16的表达。结果:手工制作的组织芯片阵列排列整齐,仅有1个组织缺失(1/625=0.16%),3个组织有折叠(3/625=0.48%),其余组织形态均好;ER、PR在乳腺癌、宫颈癌、子宫内膜癌、卵巢癌、肺癌、食管癌、胃癌等多种肿瘤中均有强弱不等的表达;P53在肺癌及不典型增生组织中阳性表达率(70%,40%)明显高于在正常支气管黏膜中的表达(0%)(P<0.05);P16蛋白在肺鳞癌中失表达率为57.9%,其中有转移组(8/13=61.5%)高于无转移组(7/17=41.2%)(P<0.05)。结论:组织芯片技术能快速、高效地对大量肿瘤组织样本进行各种检测分析,低消耗、可比性强,手工制作简单又经济,可以广泛地应用于肿瘤病理研究的各个方面;应用组织芯片技术,检测到ER,PR在多种肿瘤中均有表达,提示两者与多种肿瘤的发生有关;证明了P53基因的突变是肺癌发生的早期事件,参与了肺癌的发生与发展;P16基因在肺鳞癌中有一定的失表达率,而且与肿瘤预后相关因素中是否转移有一定相关性。

Abstract:Objective:To study the expression of various antigen in a variety of tumor tissue microarray,tissue microarray of tumor progression and tumor prognosis tissue microarray.To clear the application of tissue microarray technology in the study of tumor.Methods:Using tissue microarray and immunohistochemistry detect the expression of Estrogen Receptor(ER),Progesterone receptor PR in variety of tumor tissues,the expression of P53 in progress tissues of lung cancer,and the expression of P16 in squamous cell carcinoma of lung.Results:Hand-made tissue microarray arrays arranged in neat rows,only one organization was missed(1/625=0.16%),three organizations were folded(3/625=0.48%),the rest were all in good morphology.ER,PR in breast cancer,cervical cancer,endometrial cancer,ovarian cancer,lung cancer,esophageal cancer,gastric cancer and other tumors were all expressed,and the expression is different;The expression of P53 in lung cancer and atypical hyperplasia(70%,40%) was significantly higher than it in normal bronchial mucosa(0%)(P<0.05);The lost pression rate of P16 protein in lung squamous cell carcinoma was 57.9%,in which metastasis group(8/13=61.5%) was significantly higher than those group without metastasis(7/17=41.2%)(P<0.05).Conclusion:Tissue microarray technology can quickly and efficiently carry through various testing and analysis on a large number of tumor tissue samples,It is low-cost,strong comparability,hand-made is simple and economical,can be widely applied to various aspects of tumor pathology.Application of tissue microarray technology have detected ER,PR were expressed in a variety of tumors,suggesting that both those were related with a variety of tumors;proved P53 gene mutation is an early event in lung cancer,participated in the development of lung cancer;The certain loss of expression rate of P16 gene in squamous cell lung cancer indicated it related with tumor prognostic factors whether metastasis or not.
