肝癌细胞中IL-18表达及其调控机制的研究

摘    要:
目的探讨肝癌细胞调节IL-18生物学功能的主要分子机制。方法体外培养正常人类肝细胞株QSG-7701和原发性肝癌细胞株(MHCC97-L和HCCLM3),采用RT-PCR法,分别检测这3种细胞株中IL-18及其相关正负调节基因mRNA的表达变化。结果与2种原发性肝癌细胞株比较,正常人类肝细胞株中IL-18及其正调节基因粒细胞蛋白水解酶3(proteinase-3)mRNA表达量较高,但正调节基因caspase-1和负调节基因caspase-3 mRNA表达水平无统计学差异。结论原发性肝癌细胞中IL-18 mRNA表达减弱,可能与proteinase-3表达失衡有关。
