糖尿病动脉粥样硬化大鼠血清中二羰基化合物含量的变化及其临床意义的观察

摘    要:目的 观察糖尿病动脉粥样硬化(AS)大鼠血清中二羰基化合物的表达。方法20只GK大鼠采用左旋硝基精氨酸甲酯(L-NAME)和高脂饲料喂养法制备糖尿病AS模型(Model组),另选20只健康Wistar大鼠作为对照(NC)组。应用气相色谱-质谱联用仪(CA-MS)对大鼠血清进行分离、纯化,测定其中二羰基化合物甲基乙二醛(WIG)和3—脱氧葡萄糖醛酮(3-DG)的含量。结果Model组血清中二羰基化合物MG和3-DG;含量分别为(216.7土15.6)μmol/L和(19.5±3.8)μmol/L,与NC组(85.3±24.9)μmol/L和(8.2土2.4)μmol/L相比,含量升高(P〈0.05)。结论二羰基化合物含量升高可能是糖尿病As的发病机制之一。

Changes of serum level of dicarbonyl compounds in diabetic atherosderosis rats and i.ts clinical significance

Abstract:Objective To observe the change of the concentration of dicarbonyl compounds in serum of diabetic atherosclerosis rats. Methods Twenty Goto-Kakizaki (GK) rats were randomized into the control and model groups. The diabetic atherosclerosis rat models were produced by feeding NG-Nitro-L- arginine methyl ester (L-NAME), high fat and sucrose. Another 10 Wistar rats were taken as the controls. Gas chromatography-mass spectrometry (GC-MS) was applied to determine the concentration of dicarbonyl compounds of methylglyoxal (MG) and 3-Deoxyglucosone (3-DG) in serum. Results The concentration of MG and 3-DG was significantly higher in diabetic atherosclerosis rats (216.7± 15.6) μmol/L and (19.5±3. 8) μmol/L respectively than in the normal control group (85.3±24. 9) μmol/L and (8.25±2. 4) μmol/L respectively (P〈0.05). Conclusion The elevated levels of dicarbonyl compounds may be one of the mechanisms of diabetic atherosclerosis.
