摘    要:
本文应用体外细胞培养技术,通过测定豚鼠肺泡巨噬细胞培养液中乳酸脱氢酶活性、人胚肺二倍体成纤维细胞的DNA合成及扫描电镜观察,研究了我国有代表性的4种硬质合金粉尘的细胞毒性。结果表明,硬质合金粉尘可引起乳酸脱氢酶活性一定程度升高,和成纤维细胞DNA合成不同程度降低,以及细胞表面形态学改变,说明硬质合金粉尘具有一定的生物学作用,其细胞毒性明显低于二氧化硅粉尘。

Abstract:
The cytotoxicities of four representative hard metal dusts were studied in vitro by determining the activities of lactic dehydrogenase (LDH) in the culture medium of the alveolar macrophages of guinea pigs and ~5H-thymidine incorporation to DNA of fibroblasts. The morphology changes of the macrophages were also observed by the scanning electron microscopy. The results indicated an increase of LDH activity in case of exposing to hard metal dusts, except tungsten carbide (WC) and a decrease of DNA synthesis in two kinds of hard metal dusts in certain extent. It suggested that hard metal dusts might elicit some cytotoxicity but significantly lower than that of SiO_2.
