人类血小板抗原-1系统基因分型方法的建立及初步应用

目的建立人类血小板抗原-1系统的基因分型方法,并对本地区献血小板者进行血小板抗原-1基因分型。方法以人类基因组DNA为模板,设计特异性引物,扩增目的基因,用琼脂糖凝胶电泳观察;同时将PCR产物纯化后进行测序,将其结果进行同源性比较;再用已建立的方法对本地区的献血小板者进行人类血小板抗原-1基因分型。结果成功建立了人类血小板抗原-1基因分型方法;本地区的HPA-1a、HPA-1b基因频率分别为97.7%、2.3%。结论构建的人类血小板抗原系统-1基因分型方法结果准确,操作简单,为建立血小板血型库奠定基础。

The foundation and application of a genotyping method of the human platelet antigen 1 system

Objective To set up a genotyping means of the human platelet antigen of 1 system and study genotyping of platelet antigen 1 system of the donors who donate platelet in our region. Methods Human genomc DNA was used as template, specific primers were designed, the objective genes were amplified, and the results were observed by gel electrophoresis. The PCR product were purified and determined sequence and the results were homogeneous compared. The genotyping of platelet antigen of 1 system of the donors in our region were analysed with the means. Results We successfully founded the genotyping means of the human platelet antigen of I system, The gene frequency in our region were 97.7% for HPA - 1a and 2.3% for HPA - 1b respectively. Conclusions The genotyping means of the human platelet antigen of 1 systcm,countructed by us is simple and is exact. Which is the foundation for the platelet blood group bank.
