不同浓度纳米银对血管内皮细胞增殖作用的影响

摘    要:
目的:观察不同浓度纳米银对体外培养血管内皮细胞增殖的影响.方法:取3只5 d龄SD大鼠,分离心脏,在体外酶消化法分离血管内皮细胞并进行培养,第3代细胞用于实验.除空白对照组外,血管内皮细胞分别于含0.5,0.25,0.125,0.0625,0.03125 g/L纳米银的培养基中进行培养.24 h后倒置显微镜下观察细胞形态并进行细胞计数,采用甲基噻唑基四唑比色法和流式细胞术检测血管内皮细胞的增殖情况,并采用LDH法检测急性细胞毒性.结果:与空白对照组比较,不同浓度的纳米银组血管内皮细胞细胞数减少,纳米银浓度为0.25 g/L时,对血管内皮细胞的抑制作用最明显,其他浓度彼此间无统计学差异.不同浓度纳米银对血管内皮的增殖均有一定抑制作用,但并没有急性细胞毒性.结论:纳米银对体外培养的血管内皮细胞有明显的抑制增殖作用,其作用与纳米银的剂量有关.
