复方冬凌草甲素对肺癌SPCA-1细胞的端粒酶活性的影响

摘    要:研究复方冬凌草甲素对肺癌SPCA—l细胞株的体外抗肿瘤作用并对其作用机制进行探讨。方法:以肺癌SPCA—l细胞株为研究对象,以不同浓度的复方冬凌草甲素作用于体外培养的SPCA—l细胞,检测不同药物浓度对细胞端粒酶活性的影响。在普通及倒置显微镜下观察细胞生长状态。结果:不同浓度的药物作用一定时间后均可诱导细胞发生凋亡,流式细胞仪的检测表明细胞凋亡率在一定范围内随作用时间的延长而增加。结论:复方冬凌草甲素对SPCA—l细胞株具有明显的体外抗肿瘤作用。其抗肿瘤的机制主要表现为细胞端粒酶活性明显下调,细胞粒端酶活性的调节可能是通过间接途径实现的,凋亡途径可能参与了端粒酶活性的调控。

Abstract:Objective To investigate the mechanism of growth inhibition and the variation of telomerase activity in lung cancer SPCA 1 cells by Oridonin. Methods The variations of both morphology and proliferation of lung cancer SPCA 1 cells were observed in culture media with various concentrations of Oridonin at different time in vitro. Inhibition of lung cancer SPCA 1 cells by Oridonin was detected using MTT assay. Apoptosis and morphology variations were investigated by microscopic inspection and flow cytometry (FCM). The variation of telomerase activity was detected by sensitivity silver staining semi quantitative TRAP in order to analyze the influence of Oridonin on the characteristics of in vitro cultured lung cancer SPCA 1 cells. Result The variation of SPCA 1 cell was the lower expression of telomerase activity after treated by Oridonin, suggesting that Oridonin can inhibit the growth and proliferation of SPCA 1 cells significantly. Conclusion Oridonin can effectivtly inhibit the growth and proliferation of SPCA 1 cells. Inducing cell apoptosis and directly inducing cell necrosis are the main mechanism of Oridonin. The telomerase activity is remarkably decreased concurrently when apoptosis occurred induced by Oridonin. Oridonin can influent the telomerase activity during the treatment of SPAC 1 cells in vitro. The negative regulatory mechanism of telomerase activity may be caused by cell apoptosis.
