摘    要:目的:探讨抗肿瘤坏死因子单克隆抗体对持续缺血和缺血再灌注损伤时心肌细胞Bcl-2基因蛋白及mRNA表达的影响。方法:大白鼠随机分为7组,以活结结扎左冠状动脉的前降支,分别造成阻断冠脉血流和再灌注。以S-P免疫组化及逆转录聚合酶链反应(RT-PCR)法分别检测Bcl-2基因蛋白与mRNA的表达变化。结果:抗肿瘤坏死因子单克隆抗体处理组与持续缺血组、缺血再灌注组比较Bcl-2基因蛋白及mRNA表达明显增强。结论:抗肿瘤坏死因子单克隆抗体预处理能上调Bcl-2基因蛋白及mRNA表达。

Abstract:Objective: To study the effect of anti-TNF-mAb on the expression of protein and mRNA of Bcl-2 gene in the myocardial cells during continuous ischemia and ischemia reperfusion injury. Methods: Wistar rats were randomly divided into 7 groups. The anterior descending branches of the left coronary artery was ligated with slipknot to block the blood flow and the myocardium was reperfused. Changes of the expression of protein and mRNA of Bcl-2 gene were examined by S-P immunohistochemical staining and RT-PCR analysis respectively. Results: Compared with continuous ischemia group and ischemia reperfusion group, the expression of protein and mRNA of the Bcl-2 gene was increased significantly in the anti-TNF-mAb-administrated group. Conclusion: Pretreatment with anti-TNF-mAb could up-regulate the expression of protein and mRNA of Bcl-2 gene.
