熄风静宁颗粒对DA及5-HT激动剂引发小鼠行为改变的影响

摘    要:目的观察熄风静宁颗粒对多巴胺(DA)及5-羟色胺(5-HT)激动剂引发小鼠行为改变的影响,并测定小鼠脑内单胺类神经递质及其代谢产物,探讨熄风静宁颗粒的作用与脑内DA和5-HT神经系统的关系。方法分别用苯丙胺(Amph)、阿朴吗啡(APO)和5-羟色胺酸(5-HTP)制造小鼠多动模型、攀爬模型和头部抽动模型,观察熄风静宁颗粒对这些模型的对抗作用,单胺类神经递质及其代谢产物用高效液相-电化学法(HPLC-ECD)测定。结果熄风静宁颗粒能对抗Amph引起的小鼠多动行为、APO引起的小鼠攀爬行为和5-HTP引起的小鼠头部抽动行为。熄风静宁颗粒可提高小鼠脑内高香草酸(HVA)含量,并降低5-HT及5-羟吲哚乙酸(5-HIAA)的含量,作用趋势与阳性药氟哌啶醇相一致。结论熄风静宁颗粒可能有影响小鼠脑内DA神经及5-HT神经系统功能的作用。

Abstract:Objective To observe the influence of Xifengjingning Granules on mouse behavioral changes by dopamine(DA) and 5-hydroxytrystamine(5-HT) agonist,detect the monoaminergic neurotransmitter and metabolites in mouse brain and investigate the relationship between Xifengjingning Granules effect and brain DA and 5-HT system.Methods To establish the mouse hyperkinetic model,climbing model and head twitching model by using Amph,apomorphine(APO) and 5-HTP.The antagonisms of Xifengjingning Granules on these models were observed and the monoaminergic neurotransmitter and metabolites were detected by HPLC-ECD.Result Xifengjingning Granules had the antagonistic effect on mouse hyperkinetic behavior induced by Amph,climbing behavior induced by APO and head twitching behavior induced by 5-HTP.Xifengjingning Granules increased the content of homovanillic acid(HVA) and decreased the contents of 5-HT and 5-hydroxyindoleacetic acid(5-HIAA) in mouse brain.Xifengjingning Granules had the same effective trend with haloperidol.Conclusion Xifengjingning Granules probably influences the function of DA and 5HT system in mouse brain.
