半枝莲提取物抗人舌鳞癌SAS细胞增殖作用的研究

摘    要:目的:观察半枝莲提取的总黄酮(ESB)在体外对人舌鳞癌SAS细胞超微结构和增殖力的影响,探讨其可能的作用机制。方法:用0.1mg/mL,0.5mg/mL,2.5mg/mL和10.0mg/mLESB分别作用于SAS细胞24~72h,用四甲基偶氮唑盐(MTT)比色法检测细胞活性;透射电镜观察细胞超微结构的变化;免疫组化P—V两步法检测凋亡相关蛋白Bcl-2,Bax,Caspase-3用药前后表达变化。结果:ESB在体外对人舌鳞癌SAS细胞增殖有抑制作用,且呈时效及量效关系;透射电镜观察到典型凋亡细胞,线粒体等细胞器的形态变化与ESB的浓度和作用时间有关;用药后凋亡相关蛋白Bcl-2表达减弱,Bax,Caspase-3表达增强。结论:ESB可影响舌癌细胞的超微结构,降低细胞的增殖力从而抑制舌鳞癌SAS细胞过度生长;ESB诱导舌鳞癌细胞凋亡,可能与下调Bcl-2,上调Bax,Caspase-3等凋亡相关蛋白表达有关。
