免疫磁珠法筛选人乳牙牙髓干细胞及其培养鉴定

摘    要:
摘要:目的应用免疫磁珠法分离筛选人乳牙牙髓干细胞,并做培养鉴定。方法采用STRO-1为标记物以免疫磁珠分离
筛选人乳牙牙髓干细胞,观察细胞形态及生长情况,流式细胞仪检测细胞表型,并检测细胞体外多向分化能力。结果应
用免疫磁珠法筛选人乳牙牙髓细胞可获得乳牙牙髓干细胞,经统计学处理证明看其生长速度慢于牙髓细胞,细胞表型分
析证实CD29、CD105高表达,CD34、CD45低表达。矿化诱导和成脂诱导证实STRO-1+乳牙牙髓细胞具有干细胞多向分
化的能力。结论免疫磁珠法是有效的分离纯化人乳牙牙髓干细胞的方法。所分离的细胞具有干细胞的表型特点及多向
分化能力。

Abstract:
Objective To isolate stem cells from human exfoliated deciduous teeth(SHEDs) and identify their phenotypes and multi-lineage differentiation potential. Methods Human pulp tissue from exfoliated deciduous teeth were dissected and digested to obtain the single cell suspension.
