补骨脂-肉豆蔻不同溶剂提取物对肠易激综合征内脏高敏感大鼠的治疗作用

摘    要:目的::比较补骨脂-肉豆蔻不同溶剂提取物对肠易激综合征( IBS)内脏高敏感大鼠的治疗作用。方法:新生SD大鼠48只,随机分为6组,各8只。除正常组外,其余各组采用母子分离结合乙酸灌肠法复制内脏高敏感大鼠模型,于造模第6周起,给药组分别给予补骨脂-肉豆蔻醇水双提物、醇提物、水提物和匹维溴铵灌胃,1次/天,连续2周。观察各组大鼠结肠组织病理学变化及大鼠内脏高敏感性相关指标,并采用免疫荧光检测大鼠结肠促肾上腺皮质激素释放因子( CRF)和CRF1型受体蛋白表达。结果:补骨脂-肉豆蔻醇水双提物、醇提物、水提物均能够升高内脏高敏感性大鼠腹部回缩反射阈值( P<0.05~P<0.01),降低结肠CRF、CRF1型受体表达。结论:补骨脂-肉豆蔻不同溶剂提取物对内脏高敏感性大鼠均有一定的治疗作用,以补骨脂-肉豆蔻醇提物疗效最佳。

Abstract:Objective:To compare the therapeutic effects of different extracts from Psoralea corylifolia L and Nutmeg on the visceral hypersensitivity rats with irritable bowel syndrome( IBS) . Methods:Forty-eight newborn SD rats were randomly divided into six groups (8 rats each group). Except the normal group,the rest groups were treated with Mother-Child separation combined with acetic acid enema to establish the visceral hypersensitivity rat model. At the beginning of the sixth week after establishing model,all model rats were irrigated using the water double-extract,alcohol extract and water extract from Psoralea corylifolia L and Nutmeg,and positive medicine of pinaverium bromide once a day for 2 weeks, respectively. The pathological changes and visceral hypersensitivity-related indicators were observed in IBS rats. The expressions of CRF and CFRR1 were determined by immunofluorescence technique. Results:The water double-extract,alcohol extract and water extract from Psoralea corylifolia L and Nutmeg could enhance the reflex threshold of abdomen retraction(P<0. 05 to P<0. 01),and decrease the levels of CRF and RFR1 in colon in visceral hypersensitivity rats. Conclusions:Different extracts from Psoralea corylifolia L and Nutmeg have certain treatment effect on the visceral hypersensitivity rats,and the effect of alcohol extract is the best.
