银黄注射液及口服液中绿原酸和黄芩苷在大鼠体内的药动学比较

摘    要:目的 比较银黄注射液和银黄口服液中绿原酸和黄芩苷在大鼠体内的的药物动力学特征.方法 10只(6)大鼠随机均分成两组,分别iv银黄注射液和ig银黄口服液,用HPLC同时测定不同时间点血浆中绿原酸和黄芩苷的浓度.结果 大鼠iv银黄注射液后,血浆中能够同时检出绿原酸和黄芩苷,其中绿原酸的呈三室开放模型,黄芩苷的药-时曲线变化呈二室开放模型;大鼠ig银黄口服液后,血浆中检不出绿原酸,而黄芩苷的药-时曲线呈多峰现象,其绝对生物利用度为15.2%.结论 不同的给药途径,绿原酸和黄芩苷在大鼠体内所表现的药物动力学特征相差很大.

Abstract:OBJECTIVE To compare the pharmacokinetic parameters of chlorogenic acid and baicalin in Yinhuang injection and Yinhuang oral solution in rats. METHODS 10 male rats were randomly divided into two equal groups, which were intravenously administrated with Yinhuang injection and intragastrically administrated with Yinhuang oral solution, respectively. The concentration of chlorogenic acid and baicalin in plasma was determined simultaneously by HPLC. RESULTS The concentration of chlorogenic acid and baicalin in plasma could be determined simultaneously after intravenous administration of Yinhuang injection; the plasma concentration versus time data of chlorogenic acid followed the three open - compartment model and that of baicalin followed the two open - compartment model. The chlorogenic acid in plasma could not be determined after intragastric administration of Yinhuang oral solution and the plasma concentration versus time data of baicalin presented multiple - peak phenomenon, and the absolute bioavailability of baicalin was about 15.2%. CONCLUSION The pharmacokinetic characters of chlorogenic acid and baicalin in Yinhuang injection and Yinhuang oral solution in rats varied greatly.
