摘    要:
目的 研究慢性非细菌性前列腺炎(chronic nonbacterial prostatitis,CNP) SD大鼠前列腺组织中转化生长因子(TGF-β1)和核心蛋白聚糖(DCN)的表达,并初步探讨前列腺组织纤维化的发病机制.方法 将60只6月龄SD大鼠随机分为对照组、30 d和45 d CNP模型组各20只.CNP模型组采用去势+高剂量苯甲酸雌二醇肌注方法建立.采用双抗体夹心法检测SD大鼠前列腺组织中TGF-β1、DCN的含量.结果 ①45 d、30 d CNP模型组大鼠TGF-β1表达水平[(140.78 ±37.94)、(106.28 ±30.63) ng/L]均显著高于对照组[(13.42-±4.24)ng/L,P<0.05];且45 d CNP模型组高于30 d CNP模型组(P<0.05);②45 d、30 d模型组DCN表达水平[(947.06±114.28)、(722.96±110.66) ng/L]显著高于对照组[(63.97±15.34) ng/L,P<0.05];且45 d CNP模型组高于30 d CNP模型组(P<0.05);③TGF-β1、DCN在CNP大鼠模型前列腺组织中表达呈正相关.结论 TGF-β1、DCN可能参与CNP发病过程,在前列腺纤维化的形成中起着重要的作用.

Abstract:
Objective To investigate the expressions of transforming growth factor-beta1 and DCN in the prostatic tissue of rat models of chronic nonbacterial prostatitis (CNP),and to explore the mechanisms of CNP and its fibrosis.Methods Sixty 6 month old SD rats were randomly allocated into three groups:normal control,30 d CNP model and 45 d CNP model,the models made by castration +high dose intramuscular injection of estradiol benzoate.The expressions of TGF-β1 and DCN in the prostatic tissue were measured using enzyme-linked immuno sorbent assay (ELISA).Results ①TGF-β1 level was higher in the 45 d and 30 d CNP rat models [(140.78 ± 37.94) ng/L,(106.28 ± 30.63) ng/L] than controls [(13.42 ± 4.24) ng/L,P < 0.05],even more significantly in the 45 d than in the 30 d group.②DCN level was higher in the 45 d and 30 d CNP rat models [(947.06 ±114.28) ng/L,(722.96±110.66) ng/L] than controls[(63.97-±15.34) ng/L,P<0.05],even more significantly in the 45 d than in the 30 d group.③And the expression of TGF-β1 was positive correlated with that of DCN in the CNP rat models.Conclusions TGF-β1 and DCN may be involved in the pathogenesis of CNP,and have an important role of the formation of prostatic fibrosis.
