纳美芬对老年大鼠术后早期脑功能影响及其机制研究

摘    要:目的 探索纳美芬对老年大鼠术后早期脑功能的影响及其机制.方法 采用随机数字表法将36只20~22月龄雄性SD大鼠分为空白对照组(10组)、手术对照组(I1组)、纳美芬处理组(N组),每组12只.10组不做任何处理,I1组与N组建立大鼠剖腹探查模型,N组于手术结束时肌内注射纳美芬0.1 mg/kg.使用恐怖适应实验检测大鼠术后1d记忆能力(术前3d进行条件反射训练),采用ELISA检测大鼠术后1d脑组织促炎因子IL-1β、TNF-α浓度以及神经细胞凋亡蛋白caspase-3表达浓度. 结果 与10组比较,I1组与N组大鼠TNF-α(38.3±2.3)、(29.4±2.5) ng/L比(21.4±2.1) ng/L]、IL-13(31.6±3.3)、(21.2±2.0) ng/L比(14.2±2.9) ng/L]表达增多(P<0.01),细胞凋亡蛋白caspase-3(109±10)、(86±9) ng/L比(65±8) ng/L]表达上升(P<0.01),僵直时间下降(P<0.01).与I1组比较,N组大鼠手术后TNF-α、IL-1β表达下降(P<0.01),细胞凋亡蛋白caspase-3表达下降(P<0.01),大鼠术后僵直时间上升(P<0.01). 结论 手术可导致老年大鼠脑内炎症反应,引发神经细胞凋亡,记忆能力下降,使用纳美芬可抑制这种改变,改善手术引起的大鼠记忆能力下降.

Abstract:Objective To investigate the effects and mechanismn of nalmefene on early memory in rats.Methods Aged rats were randomly allocated to the followiug naive group(group I0),abdominal surgery alone group(group I1),intramuscular injection of nahmefene after surgery group(group N)(n=12).The concentration of IL-1β,TNF-α and caspase-3 of brain were measured by ELISA.Rats' memory was assessed in fear conditioning test.Results Compared with that of group I0,the expression of TNF-α(38.3±2.3),(29.4±2.5) ng/L vs (21.4±2.1) ng/L],IL-13(31.6±3.3),(21.2±2.0) ng/L vs (14.2±2.9) ng/L] and caspase-3 (109±10),(86±9) ng/L vs (65±8) ng/L] in group I1 and group N rats' brain were enhanced(P<0.01),the freezing time in group Ⅰ1 and group N rats were declined (P<0.01).Compared with that of group Ⅰ1,the expression of TNF-α,IL-1β in group N rats' brain were declined (P<0.01) and the freezing time in group N rats were rised (P<0.01).Conclusions Nalmefene can restrain central inflammation and cell apoptosis to diminish the memory impairment after surgery in rats.
