扶正化瘀方对大鼠肾间质纤维化肾组织基质金属蛋白酶活性调节的作用

摘    要:
目的:探讨扶正化瘀方影响肾间质纤维化大鼠肾组织基质金属蛋白酶(MMPs)活性调节的抗肾纤维化作用机制。方法:大鼠分为正常组、模型组与扶正化瘀方组,以氯化汞灌胃诱导大鼠肾间质纤维化模型,扶正化瘀方组以4g/kg扶正化瘀方灌胃。生化检测血清肾功能、肾组织羟脯氨酸(Hyp)含量;病理染色观察肾组织病理形态改变;蛋白印迹法分析肾组织MMP-13、MMP-2、膜型基质金属蛋白酶-1(MT1-MMP)及金属蛋白酶组织抑制剂(TIMP-1/2)蛋白表达;明胶酶谱法测定MMP-2活性;荧光底物反应法检测MMP-13活性。结果:与模型组比较,扶正化瘀方组明显改善大鼠血清肾功能指标,降低肾组织Hyp含量,减轻肾组织胶原沉积,降低大鼠肾组织的MMP-2、MT1-MMP、TIMP-1/2蛋白表达,降低MMP-2活性,但增加MMP-13活性。结论:扶正化瘀方抗肾间质纤维化的作用机制在于:抑制MMP-2活性,保护肾组织基底膜;抑制TIMP-1表达,提高MMP-13活性与促进病理沉积间质性胶原的降解。
