加温联合冬凌草甲素抑制肝癌细胞SMMC-7721的生长及其机制探讨

摘    要:
目的:探讨加温联合冬凌草甲素是否具有协同抗肝癌作用,以及二者最佳联合作用模式及其可能的相关作用机制,为临床热化疗治疗肝癌提供实验依据。方法:(1)MTT检测加温与冬凌草甲素合用时对体外培养的人肝癌细胞株SMMC-7721生长的抑制作用,并且评判二者联用是否有协同作用;(2)流式细胞仪检测细胞凋亡率;(3)免疫酶联吸附法定量检测肝癌细胞培养液中AFP;(4)全自动生化仪检测培养液中ALB、ALK。结果:加温与冬凌草甲素单用及合用时均能明显抑制肝癌细胞生长,引起肝癌细胞凋亡,明显降低细胞培养液中AFP值;二者联合作用强度大于单独应用组,并且不同的联合模式产生不同的作用(P<0.05)。结论:(1)加温与冬凌草甲素联合能产生明显协同抑制肝癌细胞生长的作用,作用大小与联合的模式有关。(2)二者抑制肝癌细胞生长的机制与细胞凋亡密切相关。(3)在肝癌的临床治疗中二者联合运用可能产生明显的抑制作用。

Abstract:
Objective:To investigate the synergetic effect, the best combined pattern and the possible mechanisms of the combination of hyperthermia and oridonin,which may provide useful evidence for clinic.Methods:(1)The MTT was used to estimate the inhibition effect of hyperthermia and oridonin separately and together on cultured SMMC-7721 cell in vitro. To evaluate whether the combination of hyperthermia and oridonin had a synergetic effect. (2)DNA contents of apoptotic cells and the apoptosis index were determined by flowcytometry. (3)AFP contents was determined by ELISA. (4)ALB, ALK contents were detected by biochemistry meter of the whole automatic movement.Results:The use of hyperthermia or oridonin could remarkably inhibit growth of hepatoma cell, result in apoptosis of cancer cell and reduce the value of AFP in culture supernate; The effect in associated group was greater than those in other groups; The inhibiting effect of simultaneous use of hyperthermia and oridonin on cancer cells was better than that of sequential use of the two treatments(P<0.05).Conclusion:(1)The efficiency of hyperthermia and oridonin which had significantly inhibited the growth of hepatoma cell was related to the combined model. The combined use of hyperthermia and oridonin could produce significantly synergetic effect and decrease the dosage of oridonin which was better than using any of each alone. (2)The mechanism of hyperthermia and oridonin inhibiting growth of hepatoma cell was closely related to apoptosis. (3)Combination of hyperthermia and oridonin perhaps have significant function of anti-hepatoma in clinical application.
