Abstract:Objective To study the application value of real-time fluorescence dependent helicase isothermal amplification and real-time fluorescent RT-PCR in the hand-foot-and-mouth disease.Methods The clinical data of 106 children with suspected hand-foot-and-mouth disease were retrospectively analyzed.All children were detected by real-time fluorescent RT-PCR and real-time fluorescence dependent helicase isothermal amplification.The comprehensive diagnosis results of clinical manifestations,laboratory examination,serum and etiology were taken as the clinical diagnostic gold criteria.The diagnostic results and efficacy of different detection methods were compared.Results The window period of EV universal,EV71 and CoxA16 and average time detected by real-time fluorescence dependent helicase isothermal amplification were shorter than those by real-time fluorescence RT-PCR,the diagnostic efficiency were better than those by real-time fluorescence RT-PCR,and the positive comprehensive diagnostic rate was higher than that by real-time fluorescence RT-PCR(P<0.05).Conclusion The real-time fluorescence dependent helicase isothermal amplification has the advantages of fast,high efficiency and high diagnostic efficiency in the detection of hand-foot-and-mouth disease,and which has high clinical promotion and application value.
