天麻中天麻素的提取与含量测定

目的:建立天麻素的提取工艺和含量测定方法。方法:HPLC法测定天麻素含量,采用Diamonsil C18色谱柱(4.6 mm×250 mm,5μm),流动相:乙腈-水(5∶95),检测波长220 nm,流速1.0 m L·min-1。以天麻素提取量为评价指标,正交试验法优化回流提取天麻中天麻素的工艺。结果:测定方法的专属性、精密度和回收率良好。最佳提取条件为:提取时间为60 min、提取次数3次、料液比1∶6,在此条件下天麻素提取率为20.05 mg·g-1。结论 :该工艺提取率高,含量测定方法学考察良好。

Study on Extraction and Content Determination of Gastrodin

Objective:To optimize the extraction conditions and the content determination of gastrodin.Methods :HPLC method was used for the determination of gastrodin concentration. An Diamonsil C18column(4.6 mm×250 mm,5 μm) was used with acetonitrile-water(5∶95) as mobile phase at a flow rate of1.0 m L·min-1and detection wavelength of 220 nm. The extraction conditions of reflux extraction method were optimized by orthogonal test with extract efficiency as the index. Results:The specificity,precision and recovery of the method were satisfactory. The extraction efficiency was 20.05 mg·g-1under the optimized extraction conditions as follows:extraction time 60 min for 3 times and solvent/dry material ratio was 1 ∶6.Conclusion:The extraction efficiency was high and the method for content determination was simple with good results.
