肺癌患者抗氧化体系损伤及与细胞因子相关性研究

摘    要:
目的 研究肺癌患者体内抗氧化体系损伤及与细胞因子相关性研究。方法 采用原子荧光光度计和生物化学方法分别测定肺癌患者血中硒(Se)水平,谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、脂质过氧化产物丙二醛(MDA)活性。双抗体夹心ELISA法测定血清中白介素2受体(SIL-2R)和肿瘤坏死因子(TNFα)。结果 肺癌患者血清中Se水平,GSH-Px、SOD活性明显低于对照组,而MDA明显高于对照组(P<0.001)。癌肿完全切除后,血中Se水平,GSH-Px、SOD活性明显升高,MDA、SIL-2R、TNFα水平明显下降(P<0.05;P<0.001)。直线相关分析表明,肺癌患者中Se、GSH-Px、SOD与SIL-2R、TNFα呈明显负相关(P<0.05;P<0.001)。MDA与SIL-2R、TNFα呈明显正相关(P<0.05)。结论 肺癌患者抗氧化体系严重损伤,硒、抗氧化物酶与细胞免疫关系密切,同时与肿瘤负荷有关。

Abstract:
Objective To study the impairment of antioxidative system and correlation between antioxida-tive enzymes, free radicals and SIL2R,TNFa in patients with lung cancers. Methods The activities of serum su-peroxide dismutase(SOD) ,glutathion peroxidase(GSH-Px) and malondialdehyde (MDA) were determined by biochemical analysis and selenium level was measured by atomic fluorescent spectrophotometry in 78 patients with lung cancer and 50 healthy subjects.The concentration of serum IL-2 receptor( SIL-2R) ,TNFa were examined by double-antibody sandwich ELISA. Results Serum selenium, GSH-Px, SOD in lung cancer patients were significantly lower than those in control group( P < 0.001 respectively) , while MDA level was significantly higher than that in healthy subjects. Significant difference in SIL-2R and TNFa levels was observed between lung cancer patients and control group ( P < 0.001 respectively) . After operation, the serum Se, GSH-Px and SOD were significantly increased, while serum MDA,SIL-2R and TNFa were significantly decreased. It was found that serum Se, GSH-Px, SOD were negatively correlated ( P < 0.05 and P < 0.001 respectively) and MDA was positively correlated ( P < 0.05) with SIL-2R and TNFa. Conclusion The result showed that antioxidation system of lung cancer patients was impaired and the selenium, antioxidative enzymes and SIL-2R,TNFa may work and affect each other in immuno-pathological progress of malignant tumor, and have close relation with tumor burden.
