摘 要: | ![]() 目的用PCR法检测弓形虫感染家兔血液中弓形虫基因片段。方法弓形虫RH株速殖子感染雄性家兔16只,在感染前后耳缘静脉采集血液标本,-40℃保存。用普通PCR检测血液标本中虫体DNA基因片段。结果普通PCR检测感染家兔总阳性率为45%。感染后第1周检出率仅22%,高峰期维持在感染后2~4周,检出率约为60%左右。其后检出率下降为25%左右并一直维持。结论采用血液作为样品进行弓形虫PCR检测时其最佳检测时间为感染后1~4周,其后检出率逐渐下降。

Detection of toxoplasma gondii DNA in blood from infected rabbits by PCR |

Abstract: | ![]() Objective To detect toxoplasma gondii (T. g) DNA fragments in blood from infected rabbits by multiplex polymerase chain reaction( PCR). Methods Sixteen rabbitts were infected with T. g tachyzoites by intraperitoneal injection. The samples of blood were respectively collected from the rabbits' helix vein vessels and storded at - 40℃. Toxoplasma gondii DNA was detected by PCR. Results The positive rate of all blood samples fromthe infected rabbits was 45% and that of the blood samplesin 1 week after infection only was 29%. However, it was showed that the positive rateof the blood samples was about 60% in 2 - 4 week after infection and that was the highest in all of the infection period. Then the positive rate descended and was maintained about 22% after 4 week. Conclusions The optimal time of detection of T. g DNA in blood by PCR is in 1 - 4 week after infection and the positive rate is lower after that period. |
