三七皂苷单体不同配伍对单核-内皮细胞黏附作用的影响

摘    要:目的研究三七皂苷不同单体及单体间两两配伍对人单核-内皮细胞黏附的影响及机制。方法三七皂苷单体及不同单体配伍处理内皮细胞后,加入氧化低密度脂蛋白(ox-LDL),以孟加拉玫瑰红活细胞染色法测定人单核细胞系THP-1与人脐静脉内皮细胞(HUVECs)的黏附,并用ELISA法测定内皮细胞黏附分子(ICAM-1)在HUVECs的表达水平。结果ox-LDL显著增强THP-1与HUVECs的黏附,内皮细胞经单体Rg1、Rb1处理后,明显抑制ox-LDL诱导的单核-内皮细胞黏附及ICAM-1在HUVECs的表达(P<0.05),Rg1:Rb1、Rg1:R1、Rb1:R1配伍组对单核-内皮细胞黏附的抑制作用更为显著,并能显著下调ICAM-1的表达(P<0.01)。结论(1)三七皂苷单体Rg1、Rb1为抑制黏附的有效单体,单体Rg1、Rb1、R1之间存在某种协同作用,对ox-LDL诱导的单核-内皮细胞黏附发挥更显著抑制作用。(2)单体Rg1、Rb1及有效单体配伍组合可能通过下调ICAM-1的表达抑制单核-内皮细胞黏附,从而发挥血管内皮细胞的保护作用。

Abstract:Objective To study the effect and mechanism of different combinations of sanchinoside monomers on monocyte-endotheliocyte adhesion.Methods Oxidized low density lipoprotein(ox-LDL)was added to endotheliocytes processed by sanchinoside monomers and their different combinations.Then rose bengal staining was used to detect adhesion of human THP-1 monocytes to human umbilical vein endothelial cells(HUVECs),and enzymelinked immunosorbent assay was used to detect the expression of intercellular adhesion molecule-1(ICAM-1).Results Ox-LDL could enhance adhesion of THP-1 to HUVECs.After processed by Rg1 and Rb1,monocyte-endotheliocyte adhesion and expression of ICAM-1 decreased significantly(P<0.05),especially in combination groups such as Rg1:Rb1,Rg1:R1,and Rb1:R1(P<0.01).Conclusion Rg1 and Rb1 can obviously inhibit moncyte-endotheliocyte adhesion induced by ox-LDL,especially in different combinations of Rg1,Rb1 and R1.Rg1,Rb1 and their combinations could inhibit monocyte-endotheliocyte adhesion by down-regulating the expression of ICAM-1,which might be related to its protection effect on endotheliocytes.
