Abstract: | Insulin-like growth factors I and II (IGF-I, IGF-II) were originally identified as potent mitogens and as the mediators of growth hormone action. Besides being mitogenic, however, these polypeptide growth factors play a crucial role in cell survival, and contribute to transformation and to maintenance of the malignant phenotype. Here we will discuss signaling by the IGFs, focusing specifically on the structure and function of the IGF-I receptor and the domains of this receptor responsible for distinct IGF functions: mitogenesis, transformation, and protection from apoptosis. We will also compare the structural domains of the related but functionally distinct receptor for insulin. |
