DMBA诱导建立大鼠胰腺上皮内瘤变和胰腺癌模型

摘    要:目的研究在大鼠胰腺局部种植7,12-二甲基苯并蒽(DMBA),对胰腺上皮内瘤变(PanIN)和胰腺癌形成的诱导作用,建立类似于人胰腺癌病理学特征的动物模型。方法70只雄性清洁级SD大鼠,随机分为模型组(n=40)和假手术对照组(n=30)。模型组大鼠以10mg/100g的DMBA植入胰腺内,分别于术后1月(模型1组,n=20)和2月(模型2组,n=20)处死大鼠取材,观察胰腺大体和组织病理学改变,根据PanIN标准分析比较两模型组PanIN和胰腺癌的形成率及构成比。结果术后实验观察期内三组大鼠死亡率比较无显著差异。模型1组和模型2组PanIN的形成率分别为71%(12/17)和43%(6/14),胰腺癌的形成率分别为18%(3/17)和57%(8/14)。两模型组间PanIN和胰腺癌的构成比具有显著差异,模型2组胰腺癌形成率显著高于模型1组。假手术对照组呈正常胰腺组织。结论大鼠胰腺内10mg/100g的DMBA种植,可以在1~2月内诱导形成PanIN和胰腺癌。

Abstract:Objective To evaluate the role of 7,12-dimethyl-1,2-benzanthracene (DMBA) implantation in pancreas of rats in induction of pancreatic intraepithelial neoplasia (PanIN) and pancreatic carcinoma, and establish an animal model with pathological characteristics of human pancreas carcinoma. Methods Seventy male SD rats were randomized into two groups, model group (n=40)and sham operation group (n=30).DMBA (10 mg/100 g) was implanted into pancreas in model group, and surviving rats were killed after 1 month (model group 1, n=20) and 2 months (model group 2, n=20). The gross and histopathological observations of pancreas were conducted. The incidence and constituent ratio of PanIN and pancreatic carcinoma were evaluated between these two model groups according to PanIN classification system. ResultsThere was no significant difference in the death rate among sham operation group, model group 1 and model group 2. The incidence of PanIN in model group 1 and model group 2 were 71%(12/17) and 43%(6/14), respectively, and pancreatic carcinoma 18%(3/17) and 57%(8/14), respectively. There were significant differences in the constituent ratio of PanIN and pancreatic carcinoma between two model groups, and the incidence of pancreatic carcinoma was significantly higher in model group 2 than model group 1. No neoplastic changes were observed in sham operation group. Conclusion PanIN and pancreatic carcinoma can be induced by DMBA (10 mg/100 g) implantation into rat pancreas in 1 to 2 months.
