芪黄明目胶囊对糖尿病小鼠视网膜黏附分子的作用及其机制

摘    要:目的 观察芪黄明目胶囊对糖尿病小鼠视网膜黏附分子的作用并探讨其机制.方法 KK/Upj-Ay小鼠随机分为模型组,芪黄明目高、中、低剂量组,另设C57B L/6小鼠对照组.灌胃给药3个月,观察一般情况,测定空腹血糖(FBG);光镜及电镜观察视网膜形态学变化;Real Time PCR (qPCR)和Western blot法测定视网膜细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)、E-选择素(E-selectin)和核因子-κB(NF-κB)表达.结果 芪黄明目能不同程度改善模型小鼠症状,降低FBG;改善视网膜病理损伤;降低视网膜ICAM-l、VCAM-1、E-selectin和NF-κB mRNA和蛋白的表达.结论 芪黄明目降低糖尿病小鼠视网膜黏附分子ICAM-1、VCAM-1、E-selectin的表达与下调NF-κB有关.

Abstract:Objective To investigate the effect of Qihuangmingmu Capsule(QHMM) on retinal adhesion molecule in diabetic mice and its mechanisms.Methods KK/Upj-Ay mice were randomly divided into model QHMM(low,middle,high) groups.C57BL/6 mice were selected as control group.Mice were given intragastrically for 3 months.The general appearance and fasting blood-glucose(FBG) were observed.Pathological changes of retina were observed by light microscope and electron microscope.The expression of ICAM-1,VCAM-1,E-selectin and NF-κB were examined by Real Time PCR(qPCR)and Western blot.Results QHMM could ameliorate the symptom of DM mice.decrease FBG and improve the dathological lesions of retina.The expression of ICAM-1,VCAM-1,E-selectin and NF-κB mRNA and protein were significantly increased.Conclusion QHMM can inhibit retinal adhesion molecule ICAM-1,VCAM-1,E-selectin expression in the diabetic mice,which may be related down-regulation of NF-κB.
