尖锐湿疣患者外周血中Th17细胞及IL-17的检测

目的：检测Th17细胞、IL-17在尖锐湿疣（condyloma acuminatum，CA）患者中的表达情况，探讨Th17细胞在CA发病中的作用。方法：用流式细胞仪检测36例CA患者及24例对照者外周血中Th17细胞的比例，用酶联免疫法检测血清中IL．17的蛋白表达水平，实时荧光定量PCR检测两组外周血中IL．17mRNA的表达水平。结果：CA患者组外周血中Th17细胞的比例为（0．89±0．15）％，健康对照组为（O．02±0．04）％，两组相比，差异具有统计学意义（t=6．85，P〈0．05）；CA组患者外周血中IL-17的蛋白水平为0．90ng／L，高于对照组的0．16ng／L，差异具有统计学意义（Z=6．79，P〈0．05）；CA组患者外周血中IL-17mRNA的表达量为（16．78±23．19）×10^-4，对照组IL-17mRNA的表达量为（8．54±6．10）×10^-4，差异具有统计学意义（t=2．03，P〈0．05）。结论：CA患者外周血中Th17细胞的比例升高，其细胞因子IL-17的表达同样存在异常，提示Th17细胞可能在CA的发病中起作用。

The Expression of Th17 Cells and IL-17 in Peripheral Blood from Patients with Condyloma Acuminatum

Objective：To detect the protein and mRNA expression levels of interleukin-17 and proportions of Th17 cells in peripheral blood of patients with Condyloma acuminatum（CA） ,and to analyze their clinical significance. Methods：Venous blood samples were obtained from 36 patients with CA and 24 normal human controls. Flow cytometry was used to detect the percentage of Th17 cells in peripheral blood, enzyme linked immunosorbent assay was performed to measure the plasma level of IL-17 protein, and real time fluorescence RT-PCR was performed to detect the mRNA expression level of IL-17 in peripheral blood. Results：The percentage of Th17 cells in peripheral blood was （0. 89 ± 0. 15 ）% in patients with CA, significantly higher than that in normal human controls [ （0. 02 ± 0. 04） % ]（ t = 6. 85, P 〈 0.05 ）. Compared with normal controls, there was a significant increase in the plasma level and mRNA expression level of IL-17 in peripheral blood in patients with CA（ both P 〈 0. 05）. Conclusion：There is an elevated proportion of Th17 cells and an abnormal expression of IL-17 protein and mRNA in peripheral blood from patient with CA, indicating the possible roles of IL-17 and Th17 ceils in the pathogenesis of CA.
