关节腔注射三氧对兔膝骨性关节炎病理改变的影响

摘    要:目的通过建立兔膝骨性关节炎的模型,了解膝关节腔注射三氧对骨性关节炎软骨病理改变的影响。方法成年新西兰兔40只随机分为5组,每组8只,即正常对照组(A组),模型对照组(B组),20μg/ml三氧治疗组(c组),40μg/ml三氧治疗组(D组),60μg/ml三氧治疗组(E组)。采用Videman法对B、c、D、E组家兔进行兔膝骨关节炎模型复制。造模6周后将空气1ml注入B组家兔左膝关节腔内作为对照,将20、40、60μg/ml浓度的三氧1ml分别注入C、D、E组兔左膝关节腔内,每3d1次,治疗2次后,处死动物,观察兔膝关节软骨形态学变化及进行病理积分统计。结果B组肉眼外观及病理显示关节软骨有明显破坏,病理积分较A组明显升高(P〈0.01),C、D两组病理改变较B组显示关节软骨有所修复,病理积分较B组降低(P〈0.05),E组与B组比较病理改变无明显变化,病理积分的差异无统计学意义(P〉0.05)。结论20、40μg/ml浓度的三氧关节腔注射可促进早期兔膝骨性关节炎关节软骨的修复,有利于减轻及阻止骨性关节炎的发展;而60μg/ml浓度三氧关节腔内注射对已损伤的软骨无修复作用。

Effect of intra-articular injection of ozone on pathological changes in rabbit osteoarthritis model

Abstract:Objective To observe the effect of intra-articular injection of ozone on pathological changes in rabbit osteoarthritis model. Method Forty New Zealand rabbits were divided randomly into five groups, with 8 rabbits in each group. They were control group(A), model group(B), 20 μg/ml ozone treatment group(C), 40 μg/ml ozone treatment group(D) and 60 μg/ml ozone treatment group(E). Videman's model of knee osteoarthritis were made in the rabbits of groups B,C,D and E. Ozone 1 ml of 20μg/ml,40μg/ml,60 μg/ml was injected into rabbits' knee joint cavity in groups C,D,E and air into rabbits' knee joint cavity in group B. All rabbits were killed after twice treatments. The pathological changes of articular cartilage of rabbits' knee were compared. Result The clear damage was on articular cartilage in group B. Pathological integral in group B was obvious higher than that in group A (P〈0.01). Articular cartilage was restored in group C and D partly, and their pathological integral were lower than that in group B (P〈0.05). No pathological difference were seen between group B and E(P〈0.05). Conclusion The ozone of 20 μg/ml,40 μg/ml concentration injected into articular joint cavity may prevent the development of osteoarthritis, and promote the restoration of damaged articular cartilage. The ozone of 60μg/ml concentration may not restore the damaged articular cartilage of osteoarthritis.
