Abstract:
Objective To explore the expression and clinical significance of serum microRNA-374(miR-374),miR-451 and miR-150 in papillary thyroid carcinoma tissues.Methods The wax blocks from 104 patients with papillary thyroid carcinoma who underwent radical surgery and normal para-carcinoma tissues(over 5 cm from the lesions, confirmed with cancer-free infiltration) were enrolled.The expression levels of miR-374,miR-451 and miR-150 were detected by real-time fluorescent quantitative polymerase chain reaction(qPCR).The relationship between the 3 indexes and clinicopathological parameters in patients with papillary thyroid carcinoma was analyzed.Results The relative expression levels of miR-150,miR-451 and miR-374 in papillary thyroid carcinoma tissues were significantly lower than those in para-carcinoma tissues(P<0.05).The expression levels of miR-150,miR-451 and miR-374 in patients with TNM staging at stage T3-4 and capsule invasion were significantly lower than those with TNM staging at stage T2,and without capsule invasion.The expression level of miR-451 in patients with lymph node metastasis was significantly lower than that without lymph node metastasis(P<0.05).There was no significant difference in the expression levels of miR-150,miR-451 and miR-374 among patients with different gender, age, number of lesions and tumor diameter(P>0.05).The expression of miR-150 in papillary thyroid carcinoma tissues was significantly positively correlated with miR-451 and miR-374,and the relative expression levels of miR-451 and miR-374 were significantly positively correlated(P<0.05).Conclusion The expression levels of miR-150,miR-451 and miR-374 are related to clinical staging and capsular invasion.The miR-451 is also related to lymph node metastasis.The down-regulation of the three plays roles in the development and metastasis of papillary thyroid carcinoma.
