中药银杏叶提取物联合腺病毒介导神经营养素-3基因的神经干细胞移植治疗急性脊髓损伤的实验研究

摘    要:
目的:明确经腹腔注射中药银杏叶提取物是否对腺病毒介导NT-3基因的神经干细胞移植治疗急性脊髓损伤存在干预作用。方法:采用ALLEN打击法造模成功后随机分组:即模型对照组(A)、腺病毒介导NT-3基因的神经干细胞移植组(B)、中药银杏叶提取物联合腺病毒介导NT-3基因的神经干细胞移植组(C)、单纯腹腔注射中药银杏叶提取物组(D)。于2、6、8周各组大鼠取材,进行运动功能评分、每组5只行电生理检查、免疫组织化学染色、图像分析。结果:C组8周后大鼠BBB评分为6.24±0.15;术后2周,B组和C组的皮层体感诱发电位消失,移植后8周B、C组的波形恢复,但潜伏期延长(P0.05)。C组与B组比较潜伏期明显缩短(P0.05)。术后8周C组阳性细胞主要见于脊髓室管膜区,与B组比较染色面积较大,着色较深;术后2周,各组动物皮质脊髓束均有液化灶形成,6周后B、C组皮质脊髓束损伤区逐渐有新生的神经纤维束穿过。8周后B、C组皮质脊髓束损伤区新生的神经纤维束明显增多(P0.05)。结论:中药银杏提取物联合腺病毒介导NT-3基因的神经干细胞移植能够促进损伤神经元的修复或再生,明显改善损伤脊髓的传导功能,增加皮质脊髓束损伤区的神经纤维束数目,对急性脊髓损有临床治疗作用。
