摘    要:目的:探讨不同浓度的3H-TdR细胞内照射对细胞凋亡的影响及抑制免血管平滑肌细胞增殖的可行性.方法:将不同强度的3H标记TdR掺入体外培养的免主动脉平滑肌细胞内共同培养,在不同时期用放射性自显影技术检测3H-TdR在培养的VSMC内外分布情况及聚集部位,用流式细胞学技术观察细胞凋亡的情况及对细胞长情况的影响.结果:放射性自显影图象显示,放射性物质全部分布细胞核内,细胞核外无放射性显影,流式细胞学显示在培养的前3天见细胞凋亡比例升高,而在后5天内无明显变化,细胞生长曲线显示实验组细胞增殖受到明显抑制,呈剂量依赖方式.结论:3H-TdR能顺利通过细胞膜进入细胞内,聚积于细胞核,而且能明显抑制平滑肌细胞增殖,培养早期存在明显细胞凋亡,中晚期无影响,对静止期细胞无影响,可能是同位素辐射治疗预防血管再狭窄的一种新方法.

Abstract:Objective: We attempted to study the effects of ~3H--TdR radiation intraceller in different concentration on effecting VSM- Cs apoptosis from rabbit in vitro. Methods: Arterial smooth muscle cells cultured from New Zealand whit rabbit were exposed to ~H- TdR. The effects of ~H--TdR on cell proliferation cycle were observed with flow cytometry, where~3H--TdR in VSMCs were showed with autoradiography andapoptosis ratio curve were recorded. Results: All the ~3H--TdR concentratied in cells necleous shown by the autoradiog- raphy, the VSMC proliferation were inhibited singnifitiontly on DNA synthesis and Retarded G0/G1. G1 to S transition in VSMCs. There are not too much cell apoptosis in the procedure to be found after 3 days. Conclusion: The ~3H--TdR could entrace the cells membrane easily and gathering in the cells necleous, inhibited VSMCs proliferation efficiently, and radiated litile to all around in the same time, and can be controlled. The implication of the above is that it may protect potentially arteries from restenosis effectively. securitly and con- trolled freely after coronary revascularization.
