结直肠腺癌组织中nm23-H1和MDR-1 mRNA表达及与其临床病理因素的关系

摘    要:目的 探讨nm 2 3 -H 1和MDR -1基因表达与结直肠腺癌临床病理因素的关系。方法 在结直肠腺癌组织芯片上用原位杂交法检测 12 6例结直肠腺癌组织中nm 2 3 -H 1和MDR -1mRNA的表达 ,并与其临床病理因素进行分析。结果 结直肠腺癌组织中nm 2 3 -H 1和MDR -1mRNA阳性表达率分别为 68.2 5 %和 44 .44 % ,nm 2 3 -H 1mRNA表达与患者性别和年龄无相关性 (P >0 .0 5 ) ,与肿瘤细胞分化 (P <0 .0 1)和浸润深度 (P <0 .0 5 )相关 ,与淋巴结转移密切相关 (P <0 .0 5 )。MDR -1mRNA表达与患者性别 (P >0 .0 5 )和浸润深度 (P >0 .0 5 )无相关性 ,与肿瘤细胞分化程度 (P <0 .0 5 )、患者年龄 (P <0 .0 5 )和淋巴结转移(P <0 .0 1)相关。结论 分析nm 2 3 -H 1和MDR -1mRNA表达 ,有助于了解结直肠腺癌侵袭、分化和转移的特性 ,也有助于了解和判定结直肠腺癌多药耐药的特点 ,从而为临床治疗提供有价值的参考指标。

Abstract:Objective To study the correlation between expression of nm23 H1 and MDR 1 mRNA and invasion and metastasis of colorectal adenocarcinoma.Methods By in situ hybridization and tissue array technology,the expressions of nm23 H1 and MDR 1 in 126 patients with colorectal adenocarcinoma were determined.Results The positive expression rates of nm23 H1 and MDR 1 mRNA in colorectal adenocarcinoma were 68.25% and 44.44% respectively.The expression of nm23 H1 mRNA was not significantly associated with the gender( P >0.05) and age( P >0.05),but highly associated with the differentiation degrees( P < 0.01 ),the invasive depth( P >0.05),and the metastasis of lymph nodes( P <0.005).The expression of MDR 1 mRNA was not significantly correlated to the sexuality( P >0.05) and invasive depth( P >0.05),but significant correlation of the differentiation degrees( P <0.05),age( P <0.05) and the metastasis of lymph node( P <0.01).Conclusion The expression of nm23 H1 and MDR 1 mRNA were associated with invasion,differentiation degree,metastasis in colorectal adenocarcinoma,indicating a possible index for the multiple drugs resistance and prognosis of colorectal adenocarcinoma.
