中药金安减轻顺铂化疗副作用及增加体质量的实验研究

目的探讨中药金安是否能促进顺铂诱导 C57BL/6J小鼠 Lewis肺癌凋亡.方法 荷 Lewis肺癌的 C57BL/6J小鼠分为生理盐水组、金安组、顺铂组和金安合用顺铂组,通过检测各组小鼠的体质量变化和抑瘤率,应用流式细胞仪和 TUNEL法以及提取组织 DNA并电泳等方法观察金安合用顺铂对 Lewis肺癌的治疗作用.结果 金安组体质量明显增加,顺铂组体质量先升后降,而金安合用顺铂组体质量则先升后降再回升呈"～"型.各治疗组的抑瘤率分别为 40.90%, 52.04%和 47.83%.流式细胞仪检测各组的凋亡率依次为 14.95%, 11.68%和 14.25%.TUNEL法测定各组的凋亡指数依次是(0.79± 1.60)%,(4.00± 7.15)%,(5.93± 5.96)%,(7.07± 10.15)%; DNA提取并电泳在金安合用顺铂组有 DNA Ladder出现.结论 金安能增强顺铂诱导 Lewis肺癌的凋亡作用 ,并能减轻其毒副作用.

Experimental study on Chinese medicine Jin'an on relieving side-effect of cisplatin and increasing body mass of mice

AIM:To study whether the Chinese medicine Jin'an can promote the apoptosis of Lewis lung cancer(LLC) of C57BL/6J mice induced by cisplatin. METHODS:The C57BL/6J mice with LLC were randomized into normal saline,Jin'an,cisplatin,and Jin'an+ cisplatin groups.The therapeutic effect of Jin'an combined with cisplatin for LLC was studied by detecting the changes of body mass and tumor inhibition rate,using the flow cytometer and TUNEL,extracting DNA and electrophoresis. RESULTS:The body mass of Jin'an group was obviously increased, that of Cisplatin group increased first and then decreased, and that of Jin'an+ Cisplatin group increased first, then decreased and later increased again, in a shape of "～" . The tumor inhibition rate of all groups was 40.90% , 52.04% ,and 47.83% respectively;the apoptosis rate was 14.95% ,11.68% and 14.25% respectively; the apoptosis index was(0.79± 1.60)% ,(4.00± 7.15)% , (5.93± 5.96)% , and (7.07± 10.15)% respectively. There was the appearance of DNA Ladder in the Jin'an+ cisplatin group. CONCLUSION:Jin'an is able to promote the apoptosis of LLC induced by cisplatin, and to relieve the toxin and side effect of cisplatin.
