芍药苷对大鼠全脑缺血模型的影响

摘    要:
目的:观察芍药苷对脑缺血后再灌流期间血脑屏障及神经病理改变的影响。方法:采用四血管阻断(4VO)全脑缺血再灌流损伤模型,对模型动物给予芍药苷高、中、低剂量(分别为20,10,5 mg·kg-1)静脉注射,并设假手术组,模型组,阳性对照灯盏花素组。对大鼠神经行为学,缺血区脑组织超氧化歧化酶(SOD),丙二醛(MDA)含量,透射电镜检查,血脑屏障通透性(BBB),脑组织兴奋性氨基酸(EAA)含量进行检测。结果:芍药苷对4VO完全性脑缺血模型有明显的保护作用;芍药苷可明显增加大鼠脑组织SOD含量,降低MDA含量,减轻脑缺血对脑组织造成的氧化应激损伤;芍药苷能显著改善细胞核、核周质、线粒体、髓鞘的病理改变,对脑缺血发挥保护作用的形态学基础;芍药苷能显著减轻缺血、缺氧造成的EAA含量增加,抑制EAA所导致的兴奋性神经毒性。结论:芍药苷具有显著的保护脑缺血后脑水肿、脑组织氧化应激损伤、细胞超微结构病变、血脑屏障作用,其保护机制与抑制细胞内钙离子超载、抗自由基、降低EAA含量有关。

Abstract:
Objective: To explore the effects of paeoniflorin on blood brain barrier and pathological changes in brain ischemia. Method: Mice were divided into sham operation group, model group, positive control group-Deng zhanhua tablet group and three different dose (high, middle, low-dose) groups of paeoniflorin. The neurological symptoms of rats were observed.The SOD of ischemic brain tissue, MDA BBB and EAA contents were determined.The ultrastructure on the brain cells was inspected by transmission electron microscope. Result: Paeoniflorin had the protetive effects on 4VO model of total cerebral ischemia.Paeoniflorin could obviously increase SOD content, reduce MDA content in rat brain-tissue and alleviate oxidative stress damage by cerebral ischemia on rat brain. Paeoniflorin could improve pathological changes of cell nuclear, perikaryon, mitochondria and myelin sheath, which was the morphologic basis of the protectiue effect on ischemia. Paeoniflorin could alleviate the incrense of EAA content caused by and hypoxia and inhibit the excitatory neural toxicity by EAA. Conclusion: Paeoniflorin has the protection effect on the brain edema after cerebral ischemia, the oxidative stress damage on brain tissue, the ultrastructure lesions of cells and the BBB. The protective mechanism may be related to inhibiting intracellular calcium overload, anti-free radical and reducing EAA content.
