肾透明细胞癌中FGFR1的表达及其临床意义

目的:探讨碱性成纤维细胞生长因子受体1(fibroblast growth factor receptor1,FGFR1)在肾透明细胞癌(renal clear cell carcinoma,RCCC)中的表达及其临床意义。方法:采用SP免疫组化方法检测45例RCCC组织中FGFR1的表达。同时,用原位杂交方法检测20例RCCC组织中FGFR1mR-NA的表达。结果:在RCCC中FGFR1蛋白及FGFR1mRNA表达的阳性率分别为68.9%(31/45)、80.0%(16/20),明显高于癌旁正常肾组织(P<0.01)。FGFR1蛋白表达与FGFR1mRNA的表达呈一致性(P<0.05)。FGFR1蛋白表达与病理组织学分级和临床TNM分期相关(P<0.05)。结论:FG-FR1与肾透明细胞癌的发生发展和转移密切相关,对判断预后具有一定临床意义。

Expression and clinical significance of fibroblast growth factor receptor 1 in human renal clear cell carcinoma

Objective：To investigate the expression and their clinical relationship of fibroblast growth factor receptor 1（FGFR1）in renal clear cell carcinoma（RCCC）.Methods：S-P immunohistochemical method was used to examine the expression of FGFR1 protein in 45 cases of RCCC,which had not been treated by chemotherapy and radiotherapy before surgery.And then in situ hybridization was performed to detect the mRNA level of FGFR1 gene in 20 freezing specimens.Results：Out of 45 RCCC,the positive rates of FGFR1 protein and FGFR1 mRNA were 68.9%（31/45）,80.0%（16/20）,respectively.It was found that the expression of FGFR1 was obviously higher in RCCC than in normal tissue adjacent to the tumor（P〈0.01）.The expression of FGFR1 protein was coincidence with the FGFR1 mRNA,and was significantly correlated with the histologic grade as well as the TNM stage of tumor（P〈0.05）.Conclusions：The expression of FGFR1 is closely involved in the development,growth and metastasis of RCCC and is comprehensive indexes for prognosis.
