组织工程韧带/肌腱构建中相关细胞生长因子的研究与应用

摘    要:
背景:组织工程韧带,肌腱构建已成为目前韧带修复重建外科基础研究的热点,组织工程韧带/肌腱的体外构建与体内重塑都与细胞因子有密切关系。目的:分析2001年以来部分生长因子的研究现状,总结并列举了适合组织工程韧带,肌腱构建的生长因子,并对其作用机制进行探讨。方法:分别以“韧带、肌腱、组织工程、生长因子”,“Ligament、tendon、Tissue Engineering、growth factor”为检索词,用计算机检索Pubmed数据库及重庆维普期刊全文数据库中2001年1月至2011年12月有关文章。纳入组织工程韧带和生长因子相关文献,排除与研究目的无关和内容重复者,保留33篇文献做进一步分析。结果与结论:组织工程韧带,肌腱相关生长因子的研究较多,多数生长因子都能在促进种子细胞增殖并向腱细胞方向分化、强化组织工程肌腱的生物力学活性、减少瘢痕形成等方面发挥良好作用。通过局部注射或转基因方式能使细胞有效的表达目标蛋白,应用细胞生长因子对组织工程韧带,肌腱的构建具有重要意义。

Abstract:
BACKGROUND: Construction of tissue engineering ligament/tendon is becoming a hot spot in ligament-reconstructive research recently, which is closely associated with cytokines in vitro or in vivo OBJECTIVE: To search suitable cytokines for tissue engineering ligament/tendon construction through analyzing and summarizing the current research status of growth factors since 2001, and to explore the action mechanism of these cytokines.METHODS: A computer retrieval was performed in VIP Periodical Full-Text Database and PubMed database for articles about tissue engineering ligament and relevant cytokines from January 2001 to December 2011 using the key words of "ligament, tendon, tissue engineering, growth factor" in Chinese and English, respectively. Literatures that are repetitive in the content and irrelevant with the objective were excluded. Finally, 33 literatures were involved in further analysis. RESULTS AND CONCLUSION: There are many studies about the growth factors related to tissue engineering ligament/tendon, and most of them can improve the proliferation and differentiation of seedcells into tendon cells strengthen the biomechanical activity of tissue engineering tendon, and reduce scar formation. Local injection or transgenosis can help the cells to express target proteins efficiently. Application of cell growth factors in tissue engineering ligament/tendon construction is of great significance.
