p53蛋白和PCNA在肝癌及其癌周肝组织中表达的对比研究

摘    要:
用免疫组织化学方法检测40例原发性肝癌及其癌周肝组织中突变型p53基因蛋白(mp53)和增殖细胞核抗原(PCNA)的表达.结果表明:mp53和PCNA的表达均定位于细胞核内,表达分布是弥漫、局灶、散在3种表现.其阳性表达率:在原发性肝癌(PHC)中,mp53蛋白为45.0%(18/40),PCNA为72.5%(29/40);癌周肝组织中,mp53蛋白为2.5%(1/40),PCNA为40.0%(16/40);癌周肝硬变中mp53蛋白为11.1%(1/9),PCNA为33.3%(3/9).结果提示:mp53蛋白主要是在肿瘤细胞中表达的标志物,并能反映癌周肝细胞中存在少量癌细胞的转移;PCNA可能是各种不同细胞异常增殖处于不同细胞周期阶段很有价值的标记物.mp53基因蛋白和PCNA对原发性肝癌的发生和发展可能有重要的影响.mp53蛋白可能与正常肝细胞的恶性转化有较密切的关系,涉及一些包括PCNA以及有关细胞增殖和分化的基因调控在内的其它多种因素有关.

Abstract:
The expression of mutant p53 gene (mp53) and proliferating cell nuclear antigen (PCNA) in tumor cells and its surrounding liver cells of 40 patients with primary hepatic carcinoma (PHC) were detected by immunohistochemistry. The results showed that mp53 and PCNA were located in the cellular nuclei with diffusing, patchying, and scattering patterns of distribution. The detected rate of mp53 and PCNA were 45. 0% and 72. 5% (18/40, 29/40)in tumor tissues respectively. The positive rate of mp53 and PCNA were 11. l%(l/9) and 33. 3%(3/9);2. 5% (1/40) and 40. 0%(16/40)in surrounding cirrho-tic liver tissues and cirrhotic noncirrho-tic liver respectively. The results suggested that mp53 expression wasmainly in tumor cells and may reflect the necplasnc transformation of normal liver cells. The PCNA may be a valuable marker of proliferating status of various cells. Our data suggested that mp53 gene and PCNA may have some critical effects on carcinogenesis and development cf human primary liver carcinoma. The mp53 may be closely related to the malignant transformation of normal liver cells and could be implicated PCNA and others concerning gene control of cellular proliferation and differentiation.
