丹参对大鼠烫伤早期肝脏超微结构改变的影响

摘    要:目的:观察大鼠严重烫伤早期肝脏超微结构的变化及丹参的影响作用。方法:建立30%Ⅲ度烫伤模型,88只Wistar大鼠随机分为正常对照组(A组,8只)、平衡液复苏组(B组,40只)和丹参治疗组(C组, 40只),B、C两组烫伤后立即按Parkland公式腹腔注射平衡液120ml/kg,C组在同样治疗的基础上腹腔注射丹参注射液3ml/kg,B、C两组分别于伤后3,6,12,24,48h各时相点任选两只动物肝脏,取其左外叶分两份标本分别置于95%酒精和2.5%戊二醛小瓶中固定,光镜、电镜下观察肝组织形态学变化。结果:肝组织形态学和超微结构改变在伤后6h开始出现,并进行性加重,48h最明显;丹参治疗组肝组织病理改变轻于复苏组。结论:严重烫伤早期肝组织超微结构即可发生改变,丹参治疗后肝组织超微结构改变减轻。

Abstract:Objective: To observe changes in hepatic ultra - structure in severely scalded rats at the early stage and to study the effect of salvia miltiorrhiza injection on the changes. Method: Eighty - eight Wistar rats were randomly divided into three groups. In group A, 8 healthy rats served as normal control. In group B, 40 rats were scalded and immediately treated with intra - peritoneal injection of lactated Ringer's solution (120 mg/kg) for equilibrium resuscitation. In group C, 40 rats were scalded and treated intra - peritoneally with lactated Ringer's solution (120 mg/kg) and salvia miltiorrhiza injection (3 ml/kg). At 3, 6, 12, 24 and 48 hours post injury, 2 rats from groups B and C were killed and their hepatic left lateral lobes were fixed with 95% alcohol and 2. 5% glutaraldehyde respectively. Histological changes in the liver tissue were observed under optical and electron microscopes. Result: A series of histological and ultra - structure changes in the liver were found at 6 hours post injury and became progressively more severe and reached the worst at 48 hours post injury. In group C, the pathological changes in liver tissue was less than that in group B. Conclusion: There is a series of histological and ultra - structure changes in the liver tissue of scalded rats at the early stage post injury. Salvia miltiorrhiza injection can lessen the severity of the changes.
