宽叶缬草对大鼠局灶性脑缺血后C-Fos、C-Jun表达的影响

摘    要:
目的 :探讨宽叶缬草对大鼠局灶性脑缺血后大脑海马各区 C- Fos、C- Jun表达的影响。方法 :采用线栓法制作可复流大脑中动脉闭塞大鼠模型。 4 8只雄性 Wistar大鼠随机分为正常组 ,假手术组 ,大脑中动脉闭塞组 ,生理盐水组 ,宽叶缬草治疗组。大鼠大脑中动脉闭塞后 2 h,生理盐水组、宽叶缬草治疗组分别以生理盐水及宽叶缬草酊剂灌胃 ,每 8h一次 ;术后第 5天取脑组织分别行 C- Fos、C- Jun免疫组化检测。结果 :正常组大脑海马各区 C- Fos、C- Jun阳性细胞稀少 ;大脑中动脉闭塞后海马各区均可见大量 C- Fos、C- Jun阳性细胞 ;宽叶缬草治疗组 C- Fos、C- Jun阳性细胞数密度明显低于大脑中动脉闭塞组 (P <0 .0 5 )。结论 :宽叶缬草可能通过抑制 C- Fos、C- Jun的表达 ,减轻缺血后脑组织的病理学损害。

Abstract:
Objective:To investigate the effect of valeriana officinalis var latifolia miq(VOL) on the expression of C-Fos and C-Jun in hippocampal of the rats with focal cerebral ischemia.Methods:Reversible middle cerebral artery occlusion(MCAO) rats were induced with intraluminal suture occlusion. 48 male Wistar rats were divided randomly into 5 groups: Normal group,sham operation group, MCAO group, MCAO+normal salinegroup(NS group) and MCAO+VOL group(VOL group). The drugs, such as VOL or normal saline, were adiministered throught gastrogavage at second hour after MCAO in NS group and VOL group,once every 8 hours.The rats were sacrificed on 5th day after MCAO. Then C-Fos and C-Jun immunocyto chemistry were performed. The numerial density (ND) of all positive neurons in hippocampal were measured by CMIAS image analysis system. And the data were analyzed by SPSS 10.0 statistical software.Result:C-Fos and C-Jun positive neurons in hippocampal were found after MCAO, and the ND of C-Fos and C-Jun positive neurons had no significant difference between MCAO group and NS group; The ND of C-Fos and C-Jun positive neurons in hippocampal in VOL group were less than those in MCAO group and NS group.Conclusion:VOL inhibited the expression of C-Fos and C-Jun, which could attenuate the neuron degeneration and play a brain protective role against ischemic injury.
