人胎儿卵巢组织异种移植的实验研究

摘    要:
目的:探讨人胎儿卵巢组织异种移植后的卵泡生长发育情况。方法:18只裸鼠行去势手术后分为2组:A组为对照组,B组为胎儿卵巢组织移植组,于移植1个月后开始隔天注射5U的孕马血清促性腺激素(PMSG)持续2个月,获取移植物36h前注射hCG。获取胎儿卵巢组织行光学显微镜观察并测血清雌二醇(E2)的浓度。结果:B组血清E2水平明显高于A组,差异有统计学意义(P<0.01)。移植后存活的卵巢组织中有40%~50%的原始卵泡丢失,均以原始卵泡为主,可见少量初级卵泡和次级卵泡,未见窦卵泡的生长。移植后人胎儿卵巢组织中初级卵泡和次级卵泡的比例较移植前显著增加(P<0.01)。结论:人胎儿卵巢组织移植于裸鼠的背部皮下能存活,卵泡处于良好的生长发育状态。

Abstract:
Objective:To probe into the development of follicles in the freeze-thaw human fetal ovarian tissues after xenografted. Methods: A total of 18 castrated nude mice were divided in to 2 groups : group A was as control group, while group B was xenografted ovarian tissue group. After xenografted over one month, each nude mouse was injected with 5U pregnant mare's serum gonadotropin (PMSG) every two days for two months, and injected with 5U human chorionic gonadotrophin (hCG) 36 hours before the xenografts were obtained. All the fetal ovarian tissues were observed by microscopy and the level of E_2 was determined. Results: The E_2 levels of group B was significantly higher than that of group A(P<0.01). There was 40%~50% primordial follicles disapeared in the xenografted tissue. The most predominant type of follicles were the primordial follicles, which showed a small quantity of the primary and secondary follicles, and the antral follicle was not found. The proportion of the primary and secondary follicles in posttransplantation human fetal ovarian tissues increased obviously compared with that of in pretransplantation human fetal ovarian tissues (P<0.01). Conclusion: The human fetal ovarian tissues could survive and the follicles developed well being xenograft into the back subcutaneous of nude mouse.
