胃癌细胞药物敏感性MTT法检测结果分析

目的检测胃癌细胞对化疗药物的敏感性,为胃癌化疗个体化用药提供资料和依据。方法新鲜胃癌组织制备单细胞悬液,加入常用的五种化疗药物培养48h,MTT法观察药物对胃癌细胞的杀伤作用;选择敏感药物进行化疗者作为药敏组,经验用药化疗者作为对照组,随访和比较两组治疗效果。结果不同个体胃癌细胞对化疗药物敏感性不同。药敏组和对照组中位生存期分别为32.6个月和20.1个月,两者比较,有统计学差异;2年内生存时间无统计学差异,但2年以上生存时间药敏组显著高于对照组(P<0.05)。结论用MTT法检测胃癌对化疗药物的敏感性,是筛选化疗药物和延长生存期的有效途径之一。

Analysis on the detected chemotherapeutic drug sensitivity results of gastric cancer cells by using MTT method

Objective To evaluated the sensitivity of gastric carcinoma cells to antitumor drugs,so as to provide information and basis for selecting personalized chemotherapeutic drugs of gastric carcinoma.Method Fresh samples of gastric cancer tissue were prepared single-cell suspension and separately exposed to Tax,CDDP,5-FU,ADM,MMC for 48 hours,antitumor effects of drugs was detected with MTT assay.The group using sensitive drugs for chemotherapy were taken as chemsensitivity group and traditional chemotherapy as controlled group,the detected results of two groups were followed-up and compared.Result The sensitivity to chemotherapeutic drugs was different in different cases,averged median survival rate in chemsensitivity group and controlled group was 32.6 and 20.1 months respectively:in the survival rate within 2 yr.there was no significant differences between two groups,but more than 2-year survival rate of chemsensitivity group was remarkably higher than controlled group(P< 0.05).Conclusion Chemosensitivety testing of MTT assay could constitute the prediction on applying chemotherapeutic drug and improve survival rate.
